Electing to bat after winning the toss, Nagaland were bowled out for 89 runs in 34.1 overs.
In reply, Sikkim were 136 in 62 overs at stump on Day 1, with five wickets in hand.
Nagaland has defeated Arunachal Pradesh, Mizoram and Manipur in their previous encounters.
